You can subtract the length of the packet/sentence.

That assumes the sender doesn't insert gaps between characters which is a 
reasonable assumption to make.  You can't assume the sender's bit clock is 
super accurate but that error will be down in the noise.  (unless you do 
something aggressive to get really good timing)
