In both cases, there is usually no need to set the PAPARAZZI_HOME environment variable: it is automatically set when you do a "make" command from the paparazzi3/ directory or when you use the Paparazzi Center (./paparazzi from the paparazzi3/ directory or, if you have installed paparazzi-bin, paparazzi from the menu). The only case when PAPARAZZI_HOME (and possibly PAPARAZZI_SRC) must be set is then you want to launch an application (for example sw/ground_segment/cockpit/gcs) directly from the shell.
