That's not what I observe. I have libunistring installed on a glibc system:
$ ldd /arch/x86_64-linux-gnu/gnu-inst-libunistring/0.9.10/lib/libunistring.so
linux-vdso.so.1 => (0x00007ffc2ef0a000)
libc.so.6 => /lib/x86_64-linux-gnu/libc.so.6 (0x00007f7dc351e000)
/lib64/ld-linux-x86-64.so.2 (0x00007f7dc3c69000)
and the build of Guile 2.2.7 and Guile 3.0.2 works perfectly fine:
$ ./configure --prefix=/tmp/inst CPPFLAGS=-Wall
--with-libunistring-prefix=/arch/x86_64-linux-gnu/gnu-inst-libunistring/0.9.10
...
checking for libunistring... yes
checking how to link with libunistring...
/arch/x86_64-linux-gnu/gnu-inst-libunistring/0.9.10/lib/libunistring.so
-Wl,-rpath -Wl,/arch/x86_64-linux-gnu/gnu-inst-libunistring/0.9.10/lib
checking for libunistring version... 0.9.10
...
$ make
$ make install
Anyway, if you have a problem with the 'configure' script of guile, you
should report it to bug-guile.
