Here's the org-file with padlines set to "no" up top:
#+PROPERTY: :padlines no
* Headline
  #+BEGIN_SRC c :tangle /tmp/padlines-test.c :comments link
  hello world

And the tangle file:

/* [[file:/tmp/padlines-test.org::*Headline][Headline:1]] */

hello world

/* Headline:1 ends here */
