Sure.
Look in the manual in the section "6.2.1.1 Direct Functions"
(<http://gnus.org/manual/gnus_154.html#SEC154>).
Here is an example:
(setq gnus-secondary-select-methods
'((nnml "")
(nntp "news.gmane.org"
(nntp-open-connection-function nntp-open-ssl-stream)
(nntp-port-number 563)
(nntp-address "snews.gmane.org"))
))
(The nnml-thing is, of course, not relevant).
[This works for me on Debian GNU/Linux, using openssl, I guess, and No
Gnus 0.6].
